With no one rallying to the support of the Falcon, I think we can safely remove it as a factor.
Of the two ships remaining, the Defiant is much smaller and therefore the harder target to hit as well as being more maneuverable. In addition, the multi-frequency shields on the Defiant (which we can assume are on there because the ship was designed to fight the borg, and the primary shielding against cubes was this shielding), are designed to help fight energy weapons as well as kinetic weapons. However, the Normandy's weapons system only employs disruptor torpedoes or superheated metal. Both ships are equipped with ablative armor, and the only real edge the Normandy has when comparing shielding and armor is that they possess additional Silaris armor and Guardian defense lasers.
When comparing weapons, the Normandy has the upper hand when it comes to power, but a large design flaw, at least from what I remember, is that all its weapons are forward firing. The Thanix Cannon and Torpedoes are great when facing a large foe (which is what TIM had in mind), but less capable when combating smaller foes, (See Oculus). The Defiant faces much the same problem, but at least has one torpedo tube facing rearward allowing them some defense when attacked from behind. With the maneuverability of the Defiant, they could get right under the belly of the Normandy and deliver a lethal blow. But that all depends upon...
The Crew
CO: Sisko>Shepard- If this were a ground fight, it'd be a one-handed victory for Shepard. However, he has never actually been seen commanding the ship going through evasive maneuvers and leaves it up to Joker and EDI. Speaking of which

Helmsman: Jadzia Dax>Joker>Nog- All depending on the era. Though the Joker/EDI combo is extremely potent, they simply can't compare with the experience of the Dax symbiont, which has spent the majority of its 6/7 lives flying around in space. In addition, while EDI is still confined by some AI failsafes, AI technology in the ST universe has been long accepted and the AI system aboard the Defiant is well used to its techniques.

Gunner: Worf>Garrus - Now, Worf's official position is tactical, but let's face it. Everytime Sisko or Picard says fire, Worf always pushes the buttons. And this is another example of experience. Garrus was mainly the equivalent of a marine in the Turian Military, and Worf has been flying in an I.K.S. or a U.S.S. since he joined starfleet. Garrus took over the gunnery position aboard the SSV 2 and therefore has, what, a couple weeks of experience.

Engineer: Tali>O'Brian- Tali's the Chief Engineer of her ship, O'Brian has no engineering on his ship and sits on the bridge. Need I elaborate?

Ship's layout: Defiant>Normandy- The Normandy has four decks. Now, should the unthinkable happen (Seriously, cause I love the guy) and Joker was killed from a console explosion which also fried some of EDI's systems. Tali is most likely the next most competent pilot of the group. However, she has to sprint from engineering to the elevator, get in a conversation with Garrus talking about Quarian life support systems while listening to some god-awful elevator music during an entirely too long loading period, and then dash out through an extremely long CIC to the cockpit.
Same situation: Dax is killed by a console explosion, O'Brian/Sisko gets up from their position and shoves the dead body over and takes the helm.

Overall, the Defiant should be able to eek out a victory because of its versatility and maneuverability, being able to slip under the Normandy and tear it a new one from below. However, if you've got Nog instead of Jadzia, things get a lot more close.

First off, the Defiant isn't more maneuverable than the Normandy. The Normandy has, in addition to 3 dimensional thrust vectoring antimatter thrusters, hyrdogen fueled thrusters exclusively for maneuvering. Not only that, its mass effect drive can be used to maneuver and fly silent. The Normandy also has the only stealth system in the fight. Also, only the main thanix cannon is forward facing. The normandy has additional guns that can fire 360 degrees, and the torpedoes/missiles are homing, so they are essentially 360 degrees as well. Not that any of this matters whatsoever because the distance between the ships in a feasible battle in space is so great that maneuvrability and forward facing weapons don't really count for anything.

CO: Eh, I'll give you that.

Pilot: EDI has no failsafes, Cerberus does not build AI in accordance to Alliance Law, otherwise they'd have none at all. She does have behavioral failsafes, but as seen in ME2, Joker can override them if he wishes. She's also designed from Reaper technology, making her super advanced. As for Joker himself, I'm pretty sure you recognize his awesomeness.

Gunner: But as you said, Worf is mainly a tactical officer. Garrus has experience with space combat, in Mass Effect, he remarks about the tactics of the Normandy's stealth system in space combat. He also knew the weapon schematics for the Thanix Cannon, demonstrating his knowledge of weapon systems.

Ships Layout: Obviously the large elevator wait time is game mechanics because the game is loading a new environment. The time it would realistically take to get from one floor to another probably won't have a marked effect on space combat.

Considering that the Defiant can simply fly around the Normandy at high warp speed continously firing at it, there is really no question about this particular match.

The Normandy cannot target an enemy moving at those speeds, and it's defences are not capable of handling the amount of power that the Defiant can put out.

Right, if it can A) find the Normandy and B) is maneuverable enough to stay with the Normandy.
